This protocol describes the preparation of a Tetracycline stock solution. The concentration of this stock solution is based on the weight of anhydrous Tetracycline.
Note: Typical stock solutions for this antibiotic in selectable media are made at 5mg/mL and are used at a final
concentration of 10 μg/mL for stringent plasmids to 50 μg/mL for relaxed plasmids.
Note: The preparation in ethanol allows the solution to be stored at -20°C without degrading through successive
freeze-thaw cycles.
Note: Selective solid medium prepared with this stock solution has proven selective even after 1 year of storage.
