SOB & SOC Media Recipe

Introduction
Materials
- Yeast Extract
- Tryptone
- NaCl (Sodium Chloride)
- KCl (Potassium Chloride)
- MgSO4 (Magnesium Sulfate)
Procedure
Per 1 Liter of SOB:
- Dissolve the following in ~800mL of distilled water:
5g of Yeast Extract
20g tryptone
0.584g NaCl
0.186g KCl
2.4g MgSO4
- Bring final volume to 1000mL with distilled water using a graduated cylinder
- Sterilize the media with autoclave
Making SOB into SOC:
- Per 1L of SOB, add 20mL of filter-sterilized 20% glucose solution
CRITICALDo this under the flow hood.

Make sure SOB medium is cooled to <50 C before adding glucose if it is fresh out of the autoclave.

If using 50% glucose solution, add 8mL per 1L SOB.

We commonly work with 50mL aliquots of SOB and have a 50% stock glucose solution. For easy reference, per 50mL SOB, add 400µL of 50% filter-sterilized glucose solution.
