K. Rhaeticus Agitation Test

Introduction
Materials
- Equipment
- 2 of the 150 mm petri dish
- 60 mm petri dish
- serological pipet
- 1000µL micropipet
- 1000mL pipet tip
- 10mL pipet tip
- Bacterial cellulose homemade growth kit
- HS medium (2,000mL)
- K. Rhaeticus cell culture (2,000µL)
Procedure
- Take out 2 150mm petri dishes and place them under the flow hood (Label them with initials, date, iGEM, and test 1, test 2 etc)
- Use the 1000mL (serological pipet) and pipet 750mL of HS medium into each 150 mm petri dish.
- After that add 750µL of the K.Rhaeticus cell culture to each petri dish using a 1000µL micropipet
- Set the 150 mm petri dishes onto the wave bioreactor in the 30 degree incubator (setting 4 )
- Next place the 60mm petri dish under the flow hood
- Add 5mL of HS medium into it (to create the negative control)
Don't forget to label it!
To objective of this K. Rhaeticus Agitation Test is to observe the effects of a simulated wave bioreactor on the effects of the K. rhaeticus culture and its bacterial cellulose yield.
