The Yamanaka media is a media for growing K. Rhaeticus. Other studies have shown that it is very effective for high bacterial cellulose yields from bacterial cultures. The below procedure produces 250mL of glucose (carbon source) solution and 250mL of Yamanaka media (without a carbon source), and when combined produces 500mL of Yamanaka + glucose media.
Important Notes - The "carbon source" used can be Mannitol, Glucose, Sucrose, Fructose, or Glycerol. Fructose and Glycerol have been less effective than the other carbon sources in referenced experiments. - The ideal pH for BC production is around 5.0. This can be achieved by adjusting the media pH with HCl and NaOH. - Autoclave the carbon source separate from the other ingredients to avoid a reaction that can produce toxic products.
